NM_000179.3(MSH6):c.1202A>G (p.Asp401Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D401G variant (also known as c.1202A>G), located in coding exon 4 of the MSH6 gene, results from an A to G substitution at nucleotide position 1202. The aspartic acid at codon 401 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
