This variant lies in the MSH2 gene (transcript NM_000251.3) at coding-DNA position 1333, where T is replaced by G; at the protein level this means replaces serine at residue 445 with alanine — a missense variant. Submitter rationale: The p.S445A variant (also known as c.1333T>G), located in coding exon 8 of the MSH2 gene, results from a T to G substitution at nucleotide position 1333. The serine at codon 445 is replaced by alanine, an amino acid with similar properties. In a massively parallel cell-based functional assay testing susceptibility to a DNA damaging agent, 6-thioguanine (6-TG), this variant was reported to be functionally neutral (Jia X et al.
