Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.836T>C (p.Val279Ala), citing Ambry Variant Classification Scheme 2023: The p.V279A variant (also known as c.836T>C), located in coding exon 10 of the MLH1 gene, results from a T to C substitution at nucleotide position 836. The valine at codon 279 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
