Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.1312G>C (p.Asp438His), citing Ambry Variant Classification Scheme 2023: The p.D438H variant (also known as c.1312G>C), located in coding exon 11 of the CHEK2 gene, results from a G to C substitution at nucleotide position 1312. The aspartic acid at codon 438 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
