Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.301T>C (p.Tyr101His), citing Ambry Variant Classification Scheme 2023: The p.Y101H variant (also known as c.301T>C), located in coding exon 4 of the BRCA1 gene, results from a T to C substitution at nucleotide position 301. The tyrosine at codon 101 is replaced by histidine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
