NM_000088.4(COL1A1):c.262G>T (p.Glu88Ter) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the COL1A1 gene (transcript NM_000088.4) at coding-DNA position 262, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 88 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The E88X nonsense variant in the COL1A1 gene has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The E88X variant is not observed in large population cohorts (Lek et al., 2016). Based on currently available evidence, we consider E88X to be pathogenic.
