Uncertain significance for BENTA disease; Severe combined immunodeficiency due to CARD11 deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032415.7(CARD11):c.220+1G>A, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects a donor splice site in intron 3 of the CARD11 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and likely results in a shortened protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with clinical features of CARD11-associated atopy with dominant interference of NF-kB signaling (PMID: 33202260, 35748740). ClinVar contains an entry for this variant (Variation ID: 489270). Studies have shown that disruption of this splice site results in skipping of exon 3, but is expected to preserve the integrity of the reading-frame (PMID: 33202260). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
