This variant lies in the POGZ gene (transcript NM_015100.4) at coding-DNA position 2419, where A is replaced by T; at the protein level this means converts the codon for lysine at residue 807 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The K807X variant in the POGZ gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The K807X variant is not observed in large population cohorts (Lek et al., 2016). We interpret K807X as a pathogenic variant.
