This variant lies in the TWIST1 gene (transcript NM_000474.4) at coding-DNA position 419, where C is replaced by A; at the protein level this means converts the codon for serine at residue 140 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The S140X variant in the TWIST1 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The S140X variant is not observed in large population cohorts (Lek et al., 2016). We interpret S140X as a pathogenic variant.
