Pathogenic — the classification assigned by GeneDx to NM_001282531.3(ADNP):c.2239G>T (p.Glu747Ter), citing GeneDx Variant Classification (06012015): The E747X variant in the ADNP gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function through protein truncation. The E747X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret E747X as a pathogenic variant.
