Uncertain significance — the classification assigned by GeneDx to NM_018896.5(CACNA1G):c.2476C>T (p.Gln826Ter), citing GeneDx Variant Classification (06012015): The Q826X variant in the CACNA1G gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. However, loss of function is not a known mechanism of disease for the CACNA1G gene. The Q826X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret Q826X as a variant of uncertain significance.
