NM_000534.5(PMS1):c.829C>T (p.Arg277Ter) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The R277X variant in the PMS1 gene has been reported previously as a germline variant in association with clear cell renal cell carcinoma (Dagliesh et al., 2010). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The R277X variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret R277X as a variant of uncertain significance.
