This variant lies in the ANOS1 gene (transcript NM_000216.4) at coding-DNA position 1267, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 423 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The R423X nonsense variant in the KAL1 gene has been reported previously in association with Kallmann Syndrome (Hardelin et al., 1993). This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense mediated mRNA decay. The R423X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server).
