It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ANOS1 are known to be pathogenic (PMID: 8504298, 11297579). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with Kallmann syndrome and sexual development disorders (PMID: 8504298, 15605412, 23643382, 23849776, 26862482, 28295047, 29211946). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 488697). For these reasons, this variant has been classified as Pathogenic.
