Submitter rationale: A novel pathogenic variant was detected in NF1 gene (c.4322delT, NM_001042492.2). This sequence change creates a premature translational stop signal (p.Leu1441*) in the NF1 gene. It is expected to result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Loss‐of function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). ClinVar not contains any entry for this variant but include c.4322T>G which lead to same stop in the NF1 gene and classified as pathogenic. For these reasons, this variant has been classified as Pathogenic.
