NM_000152.5(GAA):c.1099T>G (p.Trp367Gly) was classified as Likely pathogenic for Glycogen storage disease, type II by Genomenon, Inc, Genomenon, Inc, citing Genomenon Sequence Variant Interpretation Standards - Updated: GAA p.Trp367Gly (c.1099T>G) is a missense variant that changes the amino acid at codon 367 from Tryptophan to Glycine. This variant has been observed in at least one proband with a GAA-related disorder in the compound heterozygous and/or homozygous state (PMID:27629975). The presence of pathogenic/likely pathogenic missense variant(s) at the same amino acid position indicates that this residue is likely important for protein function. It is absent or not present at a significant frequency in gnomAD. In silico models predict that this variant is possibly or probably damaging. In conclusion, we classify GAA p.Trp367Gly (c.1099T>G) as a likely pathogenic variant.
