NM_022041.4(GAN):c.1145A>T (p.Glu382Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1145A>T (p.E382V) alteration is located in exon 7 (coding exon 7) of the GAN gene. This alteration results from a A to T substitution at nucleotide position 1145, causing the glutamic acid (E) at amino acid position 382 to be replaced by a valine (V). Based on data from gnomAD, the T allele has an overall frequency of <0.001% (1/251446) total alleles studied. The highest observed frequency was 0.001% (1/113724) of European (non-Finnish) alleles. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
