Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001458.5(FLNC):c.6600G>C (p.Glu2200Asp), citing Ambry Variant Classification Scheme 2023: The p.E2200D variant (also known as c.6600G>C), located in coding exon 40 of the FLNC gene, results from a G to C substitution at nucleotide position 6600. The glutamic acid at codon 2200 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
