Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_004629.2(FANCG):c.1295C>G (p.Ser432Cys), citing Ambry Variant Classification Scheme 2023: The p.S432C variant (also known as c.1295C>G), located in coding exon 10 of the FANCG gene, results from a C to G substitution at nucleotide position 1295. The serine at codon 432 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
