Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000135.4(FANCA):c.4121G>A (p.Ser1374Asn), citing Ambry Variant Classification Scheme 2023: The p.S1374N variant (also known as c.4121G>A), located in coding exon 41 of the FANCA gene, results from a G to A substitution at nucleotide position 4121. The serine at codon 1374 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
