Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004304.5(ALK):c.1274G>T (p.Cys425Phe), citing Ambry Variant Classification Scheme 2023: The p.C425F variant (also known as c.1274G>T), located in coding exon 5 of the ALK gene, results from a G to T substitution at nucleotide position 1274. The cysteine at codon 425 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
