NM_005228.5(EGFR):c.2500G>A (p.Val834Met) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V834M variant (also known as c.2500G>A), located in coding exon 21 of the EGFR gene, results from a G to A substitution at nucleotide position 2500. The valine at codon 834 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
