Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001389.5(DSCAM):c.5185+2T>A, citing Ambry Variant Classification Scheme 2023: The c.5185+2T>A intronic variant consists of a T to A substitution two nucleotides after exon 30 (coding exon 30) of the DSCAM gene. Variants that disrupt the canonical splice site are expected to result in aberrant splicing. The resulting transcript is predicted to be in-frame and is not expected to trigger nonsense-mediated mRNA decay, although direct evidence is unavailable. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
