Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.325A>G (p.Lys109Glu), citing Ambry Variant Classification Scheme 2023: The p.K109E variant (also known as c.325A>G), located in coding exon 4 of the DNAH5 gene, results from an A to G substitution at nucleotide position 325. The lysine at codon 109 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
