NM_005359.6(SMAD4):c.632C>G (p.Thr211Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome; Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T211S variant (also known as c.632C>G), located in coding exon 4 of the SMAD4 gene, results from a C to G substitution at nucleotide position 632. The threonine at codon 211 is replaced by serine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
