NM_177438.3(DICER1):c.3763A>T (p.Ser1255Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1255C variant (also known as c.3763A>T), located in coding exon 20 of the DICER1 gene, results from an A to T substitution at nucleotide position 3763. The serine at codon 1255 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
