NM_177438.3(DICER1):c.2156C>T (p.Thr719Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T719I variant (also known as c.2156C>T), located in coding exon 13 of the DICER1 gene, results from a C to T substitution at nucleotide position 2156. The threonine at codon 719 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
