Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.4181C>A (p.Thr1394Lys), citing Ambry Variant Classification Scheme 2023: The p.T1394K variant (also known as c.4181C>A), located in coding exon 21 of the DICER1 gene, results from a C to A substitution at nucleotide position 4181. The threonine at codon 1394 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
