Uncertain significance for Hereditary pancreatitis — the classification assigned by Ambry Genetics to NM_007272.3(CTRC):c.73A>T (p.Asn25Tyr), citing Ambry Variant Classification Scheme 2023: The p.N25Y variant (also known as c.73A>T), located in coding exon 2 of the CTRC gene, results from an A to T substitution at nucleotide position 73. The asparagine at codon 25 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
