Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001903.5(CTNNA1):c.649A>G (p.Asn217Asp), citing Ambry Variant Classification Scheme 2023: The p.N217D variant (also known as c.649A>G), located in coding exon 5 of the CTNNA1 gene, results from an A to G substitution at nucleotide position 649. The asparagine at codon 217 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
