Uncertain significance — the classification assigned by Ambry Genetics to NM_000099.4(CST3):c.365del (p.Phe122fs), citing Ambry Variant Classification Scheme 2023: The c.365delT (p.F122Sfs*15) alteration, located in exon 3 (coding exon 3) of the CST3 gene, consists of a deletion of one nucleotide at position 365, causing a translational frameshift with a predicted alternate stop codon after 15 amino acids. This variant is not expected to trigger nonsense-mediated mRNA decay and impacts the last 17% of the protein. The exact functional effect of this alteration is unknown. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
