NM_000535.7(PMS2):c.1595A>G (p.His532Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H532R variant (also known as c.1595A>G), located in coding exon 11 of the PMS2 gene, results from an A to G substitution at nucleotide position 1595. The histidine at codon 532 is replaced by arginine, an amino acid with highly similar properties. This alteration was detected in cohort of 711 Russian breast cancer patients (Nikitin AG et al. Front Oncol, 2020 May;10:666). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
