Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000393.5(COL5A2):c.2987G>A (p.Gly996Asp), citing Ambry Variant Classification Scheme 2023: The p.G996D variant (also known as c.2987G>A), located in coding exon 43 of the COL5A2 gene, results from a G to A substitution at nucleotide position 2987. The glycine at codon 996 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
