Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000090.4(COL3A1):c.2332G>T (p.Asp778Tyr), citing Ambry Variant Classification Scheme 2023: The p.D778Y variant (also known as c.2332G>T), located in coding exon 33 of the COL3A1 gene, results from a G to T substitution at nucleotide position 2332. The aspartic acid at codon 778 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
