Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007194.4(CHEK2):c.71G>T (p.Ser24Ile), citing Ambry Variant Classification Scheme 2023: The p.S24I variant (also known as c.71G>T), located in coding exon 1 of the CHEK2 gene, results from a G to T substitution at nucleotide position 71. The serine at codon 24 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
