Uncertain significance for Cystic fibrosis — the classification assigned by Ambry Genetics to NM_000492.4(CFTR):c.3331T>C (p.Phe1111Leu), citing Ambry Variant Classification Scheme 2023: The p.F1111L variant (also known as c.3331T>C), located in coding exon 20 of the CFTR gene, results from a T to C substitution at nucleotide position 3331. The phenylalanine at codon 1111 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
