Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004064.5(CDKN1B):c.67C>G (p.His23Asp), citing Ambry Variant Classification Scheme 2023: The p.H23D variant (also known as c.67C>G), located in coding exon 1 of the CDKN1B gene, results from a C to G substitution at nucleotide position 67. The histidine at codon 23 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
