Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000075.4(CDK4):c.818T>C (p.Leu273Pro), citing Ambry Variant Classification Scheme 2023: The p.L273P variant (also known as c.818T>C), located in coding exon 6 of the CDK4 gene, results from a T to C substitution at nucleotide position 818. The leucine at codon 273 is replaced by proline, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
