NM_000075.4(CDK4):c.91T>A (p.Phe31Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F31I variant (also known as c.91T>A), located in coding exon 1 of the CDK4 gene, results from a T to A substitution at nucleotide position 91. The phenylalanine at codon 31 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
