Uncertain significance — the classification assigned by Ambry Genetics to NM_016507.4(CDK12):c.419C>G (p.Ser140Trp), citing Ambry Variant Classification Scheme 2023: The p.S140W variant (also known as c.419C>G), located in coding exon 1 of the CDK12 gene, results from a C to G substitution at nucleotide position 419. The serine at codon 140 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
