NM_024529.5(CDC73):c.1214T>C (p.Ile405Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I405T variant (also known as c.1214T>C), located in coding exon 14 of the CDC73 gene, results from a T to C substitution at nucleotide position 1214. The isoleucine at codon 405 is replaced by threonine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
