Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000719.7(CACNA1C):c.31C>T (p.Pro11Ser), citing Ambry Variant Classification Scheme 2023: The p.P11S variant (also known as c.31C>T), located in coding exon 1 of the CACNA1C gene, results from a C to T substitution at nucleotide position 31. The proline at codon 11 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
