NM_000719.7(CACNA1C):c.4931C>T (p.Ser1644Phe) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1644F variant (also known as c.4931C>T), located in coding exon 40 of the CACNA1C gene, results from a C to T substitution at nucleotide position 4931. The serine at codon 1644 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
