NM_007294.4(BRCA1):c.419G>T (p.Ser140Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S140I variant (also known as c.419G>T), located in coding exon 5 of the BRCA1 gene, results from a G to T substitution at nucleotide position 419. The serine at codon 140 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
