NM_007294.4(BRCA1):c.1433C>T (p.Thr478Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T478I variant (also known as c.1433C>T), located in coding exon 9 of the BRCA1 gene, results from a C to T substitution at nucleotide position 1433. The threonine at codon 478 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
