Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.4118A>G (p.Glu1373Gly), citing Ambry Variant Classification Scheme 2023: The p.E1373G variant (also known as c.4118A>G), located in coding exon 10 of the BRCA1 gene, results from an A to G substitution at nucleotide position 4118. The glutamic acid at codon 1373 is replaced by glycine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
