Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000057.4(BLM):c.3352T>C (p.Phe1118Leu), citing Ambry Variant Classification Scheme 2023: The p.F1118L variant (also known as c.3352T>C), located in coding exon 16 of the BLM gene, results from a T to C substitution at nucleotide position 3352. The phenylalanine at codon 1118 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
