NM_000051.4(ATM):c.7324C>G (p.Gln2442Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q2442E variant (also known as c.7324C>G), located in coding exon 49 of the ATM gene, results from a C to G substitution at nucleotide position 7324. The glutamine at codon 2442 is replaced by glutamic acid, an amino acid with highly similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
