Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.533C>G (p.Pro178Arg), citing Ambry Variant Classification Scheme 2023: The p.P178R variant (also known as c.533C>G), located in coding exon 5 of the ATM gene, results from a C to G substitution at nucleotide position 533. The proline at codon 178 is replaced by arginine, an amino acid with dissimilar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
