NM_001367624.2(ZNF469):c.6093G>T (p.Glu2031Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2003D variant (also known as c.6009G>T), located in coding exon 2 of the ZNF469 gene, results from a G to T substitution at nucleotide position 6009. The glutamic acid at codon 2003 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
