NM_000051.4(ATM):c.56G>T (p.Arg19Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R19I variant (also known as c.56G>T), located in coding exon 1 of the ATM gene, results from a G to T substitution at nucleotide position 56. The arginine at codon 19 is replaced by isoleucine, an amino acid with similar properties. In an assay testing ATM function, this variant showed a functionally indeterminant result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
